Nicotinamide Riboside Shows Promise for Werner Syndrome Patients

A recent clinical trial conducted at Chiba University in Japan suggests that nicotinamide riboside (NR) may offer benefits for individuals with Werner syndrome (WS), a rare genetic condition characterized by accelerated aging. The study, led by Associate Professor Masaya Koshizaka, explored the impact of NR supplementation on key health markers in WS patients.
Werner syndrome, affecting approximately nine in a million people in Japan, leads to premature development of age-related conditions such as gray hair, cataracts, and diabetes. Patients also often suffer from severe skin ulcers and face early mortality due to cardiovascular issues or cancer. Currently, effective treatments are lacking.
The research, published in Aging Cell, involved a double-blind, placebo-controlled crossover trial. Participants received either NR or a placebo for 26 weeks,then switched treatments. The team monitored NAD+ levels,skin ulcer size,arterial stiffness,and kidney function.
The study found that NR supplementation considerably increased NAD+ levels in the blood. Furthermore, it improved arterial stiffness, a key indicator of cardiovascular disease risk, reduced the area of skin ulcers, and appeared to slow the decline of kidney function.No serious side effects where reported.
According to the researchers, the multiple benefits observed across different organ systems suggest that NAD+ depletion may play a fundamental role in Werner syndrome. by targeting this mechanism, NR could possibly address several major symptoms of the disease.
“Our findings suggest NR could serve as a valuable treatment option for two major symptoms, arteriosclerosis and skin ulcers, as well as for preventing kidney function decline,” saeid Dr. Koshizaka.
