Columbia University’s Alzheimer’s Research Center has initiated a new phase of studies aimed at uncovering the underlying causes of Alzheimer’s disease, according to a report by ABC7 New York. The research, led by physicians and scientists at the center, focuses on identifying biological markers and environmental factors that may contribute to the progressive neurodegenerative condition.
The initiative builds on decades of work by the center, which has been a key player in Alzheimer’s research since the 1980s. The latest efforts involve a multi-disciplinary approach combining genetic analysis, brain imaging, and longitudinal patient studies. Researchers are particularly examining the role of amyloid-beta plaques and tau protein tangles, which are hallmark features of the disease, as well as potential links to vascular health and immune system responses.
Dr. Maria Alvarez, a neurologist and co-principal investigator on the project, emphasized the importance of understanding the disease’s origins. “Alzheimer’s is not a single condition but a spectrum of pathologies,” Alvarez said in a statement provided to ABC7 New York. “By pinpointing the earliest biological changes, we hope to develop interventions that can slow or halt the disease before symptoms emerge.”
The research is being conducted in collaboration with the National Institute on Aging (NIA), which has allocated $25 million in funding for the initiative through 2028. The NIA’s involvement underscores the project’s alignment with federal priorities to advance Alzheimer’s prevention and treatment, as outlined in the 2021 National Alzheimer’s Plan.
One of the study’s key components is a 10-year observational trial tracking 1,500 participants aged 60 and older. The cohort includes individuals with varying levels of cognitive impairment, from mild memory concerns to early-stage Alzheimer’s. Researchers are using advanced MRI scans and cerebrospinal fluid analysis to detect early changes in brain structure and chemistry.
Early findings, though preliminary, suggest a potential correlation between vascular health and Alzheimer’s progression. A separate study published in the Journal of Neurology in July 2026 found that individuals with untreated hypertension were 40% more likely to develop Alzheimer’s-related brain changes compared to those with controlled blood pressure. While the Columbia team has not yet released specific data, they have noted that vascular risk factors are a “critical area of investigation.”
The center is also exploring the role of gut microbiota in neurodegenerative diseases. A pilot study involving 50 participants found that certain bacterial strains were more prevalent in individuals with early-stage Alzheimer’s, though the significance of this association remains unclear. “We’re still in the exploratory phase,” said Dr. James Carter, a microbiologist involved in the research. “But the gut-brain axis is a promising avenue for future therapies.”
Public health experts have welcomed the research but cautioned against overinterpreting early results. Dr. Linda Nguyen, a geriatrician at the Mayo Clinic who was not involved in the study, noted that Alzheimer’s research often faces challenges in translating laboratory findings to clinical practice. “These studies are essential, but they require years of validation before they can inform treatment guidelines,” Nguyen said.
The Columbia team has also partnered with pharmaceutical companies to test experimental drugs targeting amyloid plaques. One such drug, LUM-123, showed mixed results in a Phase II trial, with some patients experiencing slowed cognitive decline but others reporting severe side effects. The center’s researchers are analyzing data from this trial to refine dosing strategies and patient selection criteria.
Despite the complexity of Alzheimer’s, the center remains optimistic about the potential for breakthroughs. “We’re at a critical juncture,” said Dr. Alvarez. “Every discovery, even small ones, brings us closer to preventing or curing this disease.”
The research is expected to culminate in a comprehensive report by 2028, which will be submitted to the NIA for review. Until then, the team plans to publish periodic updates through peer-reviewed journals and public health forums.
