Imatinib Dosage Changes impact CML Treatment Response

⁢updated June 18, 2025

A new study examines how changes in imatinib dosage affect treatment outcomes for patients with chronic myeloid leukemia (CML). The research, leveraging real-world data, reveals that dosage modifications, including reductions and interruptions, are common and can influence a patient’s response⁤ to therapy.

The findings ⁣will be presented at the Oncology Pharmacists Connect (OPC) meeting in Austin, Texas, from June 19-20. The data was initially shared at the American Society of ⁣Clinical Oncology (ASCO) ⁣Annual Meeting in Chicago,Illinois.

Researchers analyzed electronic health records from ⁣a large health system, focusing on patients receiving imatinib (Gleevec) as a first-line ⁢treatment ‍for CML. The study aimed to⁢ determine the prevalence of dosage modifications and their impact on achieving molecular response milestones.

The study‍ found that nearly half of CML patients ⁣experienced at least one dosage modification ⁣within the first year of imatinib therapy. These modifications were linked to a lower probability of ‍reaching the ⁣six-month molecular response milestone.‍ The research underscores the importance of careful monitoring and further ‍investigation into the long-term effects ‍of ⁢dose adjustments in real-world CML treatment.

The retrospective study ‍included 118 adult⁣ patients diagnosed with ⁣CML who received imatinib as their initial ⁢treatment⁢ between‍ 2008 and 2023. ⁤The median age was 61, with a majority ⁣being women (55%). The⁣ patient population ‍represented‍ diverse racial and ethnic backgrounds.

Of the patients studied, 50% (59 individuals) experienced a total of 112 dosage modification events within the first year. These events included dose reductions (26%), dosage interruptions (46%), and discontinuations (25%). more than half of these modifications occurred ⁣within the first six ⁤months of treatment. Toxicities ⁣prompted dosage modifications in 66% of patients, while a small percentage (3%) started treatment⁢ at a reduced
