Personalized Vaccine Shows promise in Treating Kidney Cancer

A phase 1 clinical trial at the dana-Farber Cancer Institute indicates a personalized cancer vaccine spurred a successful immune response in all nine patients with stage III or IV clear cell renal cell carcinoma, a type of kidney cancer. The personalized cancer vaccine, administered post-surgery, is designed to stimulate the body’s immune system to identify and eliminate any remaining cancer cells. After a median of nearly 35 months, all patients remained cancer-free.
Dr. Toni Choueiri, director of the Lank Center for Genitourinary Cancer at Dana-Farber and co-principal investigator, expressed enthusiasm about the positive results observed in all nine kidney cancer patients. The study, he said, reflects a strong collaboration between the NeoVax team, the Broad Institute of MIT and Harvard, and Dana-Farber’s Lank Center.
the personalized cancer vaccines leverage neovax vaccine technology, developed by dr. The team extracts molecular features,called neoantigens,from the patient’s tumor to differentiate cancer cells from normal ones. predictive algorithms then select neoantigens likely to trigger an immune response, which are incorporated into the vaccine.
Dr. David A. Braun, a medical oncologist at Yale Cancer Center, emphasized that this approach differs significantly from previous kidney cancer vaccine attempts. The vaccine targets elements unique to the cancer, enabling the immune system to specifically target the cancer. The study demonstrated the potential for long-lasting immune responses.
While some patients experienced mild side effects like local reactions at the injection site or flu-like symptoms, no serious adverse events were reported. according to Dr. choueiri, the vaccine’s neoantigens help direct immune responses toward cancer cells, enhancing efficacy and minimizing off-target toxicity.
The research team noted that the vaccine induced an immune response within three weeks, with T cell counts increasing significantly and remaining elevated for up to three years. In vitro studies confirmed the vaccine-induced T cells actively targeted the patients’ tumor cells.
“We observed a rapid, substantial, and durable expansion of new T cell clones related to the vaccine,” said Dr.Patrick Ott, director of the Center for Cancer Vaccines at Dana-Farber.
What’s next
Larger clinical trials are necessary to validate the vaccine’s effectiveness and explore its full potential as a kidney cancer treatment. A multicenter international study is underway,evaluating a similar neoantigen-targeting personalized cancer vaccine in combination with pembrolizumab immunotherapy (NCT06307431).
