Researchers at the University of Waterloo’s School of Pharmacy have proposed a new therapeutic strategy for treating Alzheimer’s disease by combining established anti-amyloid antibody medications with small molecule compounds derived from natural micronutrients.
The study, led by Professor Praveen Nekkar Rao, suggests that a dual approach targeting amyloid-beta protein aggregation can increase treatment efficacy while potentially reducing the frequency and severity of complications associated with antibody therapies.
Targeting Amyloid-Beta Aggregation
Alzheimer’s disease is a progressive neurodegenerative disorder characterized by memory loss and cognitive decline. A central part of the disease’s pathology involves amyloid-beta peptide aggregates, which form toxic plaques in the brain.
These plaques disrupt neuronal function and eventually lead to widespread neuronal death. While many existing treatments only alleviate the symptoms of the disease, they generally do not halt its progression.
Anti-amyloid monoclonal antibodies, such as Lecanemab and Aducanumab, were developed to directly target these amyloid-beta plaques. However, these treatments carry significant risks of adverse effects, including microbleeds and cerebral edema.
The Role of Natural Small Molecules
The University of Waterloo team investigated the potential of pairing these antibody treatments with natural small molecule inhibitors that possess anti-amyloid properties. Specifically, the research focused on curcumin and resveratrol.

- Curcumin is a bioactive compound derived from turmeric.
Both compounds have been documented to possess antioxidant and anti-inflammatory capabilities, and they are known to interfere with the formation of amyloid fibrils.
Potential Benefits of Combination Therapy
The goal of this combination approach is to use small molecules to enhance the efficacy of antibodies at lower dosages. By increasing the effectiveness of the medication through this pairing, researchers believe it may be possible to mitigate the serious side effects associated with high-dose antibody therapies.
By utilizing these micronutrient-derived molecules, the treatment aims to double down on the fight against Alzheimer’s disease by attacking the protein aggregates through two different mechanisms simultaneously.
