Omega-6 Fatty Acid’s Role: Arachidonic Acid Linked to Lower Bipolar Risk

A new study published in Biological Psychiatry suggests a connection between a genetic predisposition for higher levels of arachidonic acid, an omega-6 polyunsaturated fatty acid, and a reduced risk of bipolar disorder. Arachidonic acid is commonly found in foods like eggs, poultry, and seafood. The research opens doors for possible lifestyle and dietary changes to address the condition.
Bipolar disorder, characterized by recurring episodes of mania and depression, is a debilitating mood disorder. While the exact causes remain unclear, previous research indicates a strong hereditary component. The study’s findings highlight a potential link between bipolar disorder and altered metabolite levels, suggesting that circulating metabolites play a significant role in the development of bipolar disorder and other psychiatric conditions.
David Stacey, PhD, lead investigator at the Australian Centre for Precision Health, stated that identifying metabolites with causal roles in bipolar disorder could highlight potential lifestyle or dietary interventions. His team used Mendelian randomization to analyse 913 metabolites and found 33 associated with bipolar disorder, most of which were lipids.
the researchers also discovered that a bipolar disorder risk gene cluster (FADS1/2/3), which influences lipid metabolism, mediated the relationship between bipolar disorder and levels of arachidonic acid and other metabolites.
John Krystal, MD, editor of Biological Psychiatry, noted the study provides a step forward in developing blood biomarkers for bipolar disorder risk, especially in patients with gene variations in the FADS1/2/3 cluster.He said arachidonic acid contributes to the health of cell membranes.
Intriguingly, we observed a pattern whereby a genetic propensity to higher levels of lipids containing an arachidonic acid fatty acid side chain was associated with a lower risk of bipolar disorder, while the inverse was true of lipids containing a linoleic acid side chain,” Dr. Stacey said.
Arachidonic acid, present in human milk, is considered vital for infant brain development and is often added to infant formula. It may influence bipolar disorder risk through neurodevelopmental pathways, aligning with current views of bipolar disorder as a neurodevelopmental condition. Dietary sources include meat, seafood, nuts, seeds, and oils.

Stacey suggests further research is needed to determine the preventive or therapeutic value of arachidonic acid supplements, particularly for those with compromised synthesizing pathways or poor dietary intake. He also emphasized the potential for early life nutrition interventions to ensure adequate intake of arachidonic acid and other polyunsaturated fatty acids for optimal brain development, potentially reducing bipolar disorder risk.
