Jiří Lábus sa narodil 26. januára 1950 v Prahe. Absolvoval gymnázium, ale už od piatich rokov vedel, že che byť hercom, hoci nikto z rodičov sa tejto profesii nevenoval. Otec bol architekt, mama zdravotná sestra. „Od detstva som si šiel za tým, chodil som do divadiel, najskôr do bábkových, potom aj do klasických. Na gymnáziu bol pre mňa celý týždeň vždy príšerný, ale tešil som sa na sobotu, že pôjdem niekam do divadla,” povedal v rozhovore pre DenníkN a pokračoval: „Čo sa týka herectva, jednoducho ma bavilo sa predvádzať. Už na základnej škole som miloval, keď som bol stredobodom pozornosti. Bol som schopný sa zosmiešniť strašným spôsobom, aby sa mi celá trieda smiala. Rodičia z toho šaleli.”
Sen sa mu aj splnil, po maturite nastúpil na DAMU v Prahe a v roku 1973 získal diplom. Po škole mu šéf divadla Ypsilon Jan Schmid ponúkol angažmán vo svojom súbore, ktorého domovom bol v tom čase Liberec. Divadlo sa v roku 1978 presťahovalo do Prahy a Jiří Lábus je mu verný dodnes.
