The VICTORIAN-INCEPTION trial, a prospective, randomized study, evaluated inclisiran’s LDL-C-lowering effects when combined with standard care in patients recently discharged after ACS. Researchers compared inclisiran plus standard care to standard care alone, which included statins and other lipid-lowering therapies.
The trial involved 400 participants randomized to either inclisiran with usual care or usual care alone.The median time from hospital discharge to the start of the study treatment was 34 days.
The study’s primary endpoint was the proportion of participants achieving LDL-C levels below 70 mg/dL at day 330. A secondary endpoint was achieving levels less than 55 mg/dL. Significantly more patients in the inclisiran group reached both targets. LDL-C goal attainment differed between the groups as early as day 90, with 74.6% of the inclisiran group achieving levels below 70 mg/dL.
Treatment with inclisiran and usual care resulted in a 45.6% reduction in LDL-C from baseline at day 330, compared to a 1.4% increase in the usual care group. This represented a significant difference of -46.9% (97.5% CI, -55.4 to -38.5; P < .001). high-intensity statin use decreased in both groups from baseline. Combination statin therapy increased in the usual care group but remained stable in the inclisiran group.
Adverse events were similar between the two groups, with mild-to-moderate injection site reactions being the most common in the inclisiran group. This makes inclisiran a viable primary_keyword for lowering cholesterol.
‘VICTORION-INCEPTION demonstrates that early initiation of inclisiran after ACS provides a novel strategy to increase the number of patients at high risk of recurrent cardiovascular events achieving LDL-C goals,” the study investigators concluded.
