Medical research into pancreatic cancer saw significant progress in 2025, focusing on improving early detection, overcoming treatment resistance, and developing new vaccine therapies. These advancements address a disease that remains the third leading cause of cancer-related death in the United States and is the only major cancer with a five-year survival rate below 20%.

Much of the difficulty in treating pancreatic cancer stems from the timing of diagnosis. Approximately 80% of cases are identified only when the disease has reached its most advanced stages, meaning the tumor either cannot be surgically removed or has already metastasized to other organs, such as the lungs or liver.

Beyond the timing of detection, researchers have long struggled with the biological makeup of the tumors. About 90% of pancreatic cancers are caused by a genetic mutation known as KRAS, which was historically categorized as undruggable because scientists could not find a way to latch drugs onto the target.

Advancements in Immunotherapy and the Tumor Microenvironment

Immunotherapy has historically been ineffective against pancreatic cancer because the tumors do not exist in isolation. Instead, they are surrounded by a protective microenvironment consisting of other cell types that form a physical and chemical barrier, preventing immunotherapy drugs from reaching the cancer cells.

To combat this, researchers at MD Anderson are working to block the specific chemical signals that maintain this barrier. New methods are being developed to modify and strengthen immune cells before reinserting them into the patient’s body to target the cancer directly.
